Correlation between 405G/C gene polymorphism of vascular endothelial growth factor and acute myocardial infarction

  • 目的:分析血管内皮生长因子(VEGF)405G/C基因多态性与急性心肌梗死(AMI)的相关性。方法:选取我院2018年1月—2019年12月收治的124例AMI患者设为AMI组。同时选取120例体检的正常健康人群作为对照组。采集血样提取DNA,利用聚合酶链式反应(PCR)、酶切反应及琼脂糖凝胶电泳检测VEGF基因405G/C位点多态性;测定VEGF和心肌损伤标志物水平。Pearson相关性分析评估患者VEGF浓度与心肌指标的关系。结果:AMI组的VEGF基因405G/C位点的CC、GC和GG基因型频率分别为54.84%、25.81%和19.35%;C等位基因和G等位基因频率分别为67.74%和32.26%。和对照组比较,AMI患者CC基因型显著增加,而GG基因型明显减少,与此同时等位基因C频率增加,而等位基因G频率则减少(P<0.05),C等位基因突变患者AMI风险增加(OR=1.998,95%CI1.383~2.884)。AMI患者血清TC、LDL-C、NT-proBNP、CK-MB、cTnI及VEGF水平明显高于对照组,HDL-C含量显著降低(P<0.05)。VEGF基因405G/C位点CC基因型的AMI患者血清VEGF水平显著低于GC、GG基因型患者(P<0.05)。Pearson相关性分析发现,AMI患者VEGF水平与心肌损伤标志物NT-proBNP、CK-MB、cTnI含量均呈负相关(P<0.05)。结论:VEGF+405G/C基因多态性与AMI的发生有关,VEGF(+405C)等位基因可能是AMI的易感基因,VEGF水平与心肌损伤程度负相关。
