Correlation between 405G/C gene polymorphism of vascular endothelial growth factor and acute myocardial infarction

摘要: 目的:分析血管内皮生长因子(VEGF)405G/C基因多态性与急性心肌梗死(AMI)的相关性。方法:选取我院2018年1月—2019年12月收治的124例AMI患者设为AMI组。同时选取120例体检的正常健康人群作为对照组。采集血样提取DNA,利用聚合酶链式反应(PCR)、酶切反应及琼脂糖凝胶电泳检测VEGF基因405G/C位点多态性;测定VEGF和心肌损伤标志物水平。Pearson相关性分析评估患者VEGF浓度与心肌指标的关系。结果:AMI组的VEGF基因405G/C位点的CC、GC和GG基因型频率分别为54.84%、25.81%和19.35%;C等位基因和G等位基因频率分别为67.74%和32.26%。和对照组比较,AMI患者CC基因型显著增加,而GG基因型明显减少,与此同时等位基因C频率增加,而等位基因G频率则减少(P<0.05),C等位基因突变患者AMI风险增加(OR=1.998,95%CI1.383~2.884)。AMI患者血清TC、LDL-C、NT-proBNP、CK-MB、cTnI及VEGF水平明显高于对照组,HDL-C含量显著降低(P<0.05)。VEGF基因405G/C位点CC基因型的AMI患者血清VEGF水平显著低于GC、GG基因型患者(P<0.05)。Pearson相关性分析发现,AMI患者VEGF水平与心肌损伤标志物NT-proBNP、CK-MB、cTnI含量均呈负相关(P<0.05)。结论:VEGF+405G/C基因多态性与AMI的发生有关,VEGF(+405C)等位基因可能是AMI的易感基因,VEGF水平与心肌损伤程度负相关。Abstract: Objective: To analyze the correlation between 405 G/C gene polymorphism of vascular endothelial growth factor(VEGF) and acute myocardial infarction(AMI).Methods: The 124 AMI patients admitted to our hospital were selected as AMI group from January 2018 to December 2019. At the same time, 120 normal healthy people underwent physical examination were selected as the control group. Blood samples were collected to extract genomic DNA, and the polymorphism of VEGF gene 405 G/C site was detected by polymerase chain reaction(PCR), enzyme digestion reaction and agar-gel electrophoresis. The levels of VEGF and myocardial injury markers were also determined. Pearson correlation analysis was used to evaluate the relationship between VEGF concentration and myocardial index.Results: CC, GC and GG genotype frequencies of VEGF gene 405 G/C in AMI group were 54.84%, 25.81% and 19.35%, respectively. Allele C and G frequencies of VEGF gene 405 G/C were 67.74% and 32.26%, respectively. Compared to control group, CC genotype and allele C frequencies of AMI patients were significantly higher, and GG genotype and allele G frequencies were significantly reduced with the statistical difference(P<0.05). The risk of AMI in patients with C allele mutation was increased(OR=1.998, 95%CI 1.383-2.884). Serum TC, LDL-C, NT-proBNP, CK-MB, cTnI and VEGF levels in AMI group were significantly higher than that in the control group, and the content of HDL-C was significantly lower(P<0.05). Serum VEGF level in AMI patients with CC genotype of VEGF gene 405 G/C locus was significantly lower than those with GC and GG genotypes(P<0.05). Pearson correlation analysis found that VEGF level in AMI patients was negatively correlated with the contents of myocardial injury markers, such as NT-proBNP, CK-MB, and cTnI(P<0.05).Conclusion: The polymorphism of VEGF+405 G/C gene was related to the occurrence of AMI. The allele of VEGF(+405 C) might be the susceptible gene of AMI, and the level of VEGF was negatively correlated with the degree of myocardial injury.
