白细胞介素-17对大鼠心肌梗死后室性心律失常的影响

Effect of IL-17 expression on ventricular arrhythmias in post-myocardial infarction rats

  • 目的:探讨大鼠心肌梗死(MI)后白细胞介素-17(IL-17)在室性心律失常发生中的作用。方法:采用液氮冷冻法建立大鼠MI模型共138只,随机分为MI对照组(MI组)、小鼠同型IgG组(IgG组)和IL-17抗体组。每组又随机分为3个亚组:2周组、4周组及8周组。另设假手术组。假手术组和MI组分别腹腔注射0.8mlPBS溶液,并注入0.2ml空气促使液体分散到腹腔内;IgG组、IL-17抗体组分别注射小鼠同型IgG1+PBS溶液(100μg IgG1溶于0.8ml PBS,同样注入0.2ml空气)、小鼠抗人IL-17单克隆抗体+PBS溶液(100μg IL-17抗体溶于0.8ml PBS溶液,同样注入0.2ml空气)。每隔1d用药1次,连续用药5次。运用程序电刺激方法,观察各组室性心律失常的诱发率,同时用Western blot和激光共聚焦显微镜检测IL-17蛋白表达水平。结果:与假手术组比较,MI组大鼠心肌组织IL-17表达显著增加(P<0.05),室性心律失常诱发率亦明显增高(P<0.05);与MI组比较,IL-17抗体组IL-17表达显著减少(P<0.05),室性心律失常诱发率亦明显降低(P<0.05)。大鼠心肌IL-17的表达水平和室性心律失常诱发率之间呈正相关性(r=0.88,P<0.05),但IgG组与MI组比较差异无统计学意义(P>0.05)。结论:MI后大鼠心肌组织IL-17表达明显增加,大量IL-17表达与MI后室性心律失常的发生密切相关。IL-17的大量表达可能对MI后室性心率失常折返环的形成有诱导或促进作用。
