Mechanism of connective tissue growth factor enhance Caspase-3 activity to regulate ox-LDL induced THP-1 apoptosis

摘要: 目的:研究动脉粥样硬化过程中结缔组织生长因子 (CTGF) 在巨噬细胞凋亡过程中发挥的作用及机制。方法:对人巨噬细胞 (THP-1) 分组进行干预, 采用SiRNA基因沉默方法, 对CTGF基因进行靶向沉默, 用细胞流式、实时定量、蛋白印迹及酶活性检测等技术, 阐明CTGF在氧化性低密度脂蛋白 (ox-LDL) 诱导THP-1凋亡过程中起的作用及机制。结果:CTGF在ox-LDL刺激THP-1细胞组高表达, 而CTGF靶向性SiRNA可以降低其表达;沉默CTGF后, THP-1凋亡率明显降低, 并且与Caspase-3酶活性呈正相关。结论:内源性的CTGF可以通过增强Capsase-3酶活性来促进THP-1的凋亡。Abstract: Objective: To research the role and mechanism of connective tissue growth factor (CTGF) during process of the macrophage apoptosis.Method: We stimulated the different group of the human macrophage (THP-1).We used the RNA interfering technology to influence gene of CTGF, and observed the role and mechanism of CTGF during process of the THP-1apoptosis, which were detected by flow cytometry, PCR, Western blot, and ELISA.Result: Ox-LDL induced the high expression of CTGF in THP-1cells, and the SiRNA of CTGF decreased the expression of CTGF, this effect also influenced the apoptosis rate of THP-1cells.In addition, the activation of Caspase-3also changed with the SiRNA of CTGF.Conclusion: Endogenous CTGF can enhance the activity of Caspase-3to induce the THP-1cells apoptosis.
