Correlation between BMP4 and inflammation in the myocardial,arteries and adipose tissue of obese mice

摘要: 目的:探讨肥胖老鼠体内心脏、动脉与脂肪组织中骨形态发生蛋白4(BMP4)的表达及其与炎症程度的相关性。方法:将ob/ob小鼠分为实验组与干扰组,以C57BL/6小鼠为对照组。每组各8只,普通饮食喂养,干扰组尾静脉注射BMP4shRNA腺病毒,每周1次,持续12周。12周后心脏采血处死,取心肌组织、腹主动脉及腹部脂肪组织。免疫组织化学染色定位半定量BMP4的表达水平。PCR荧光定量和免疫蛋白印迹法检测BMP4、白细胞介素(IL)-1β和IL-9mRNA与蛋白的表达水平。结果:实验组BMP4、IL-1β和IL-9的mRNA与蛋白在心肌组织和动脉血管的表达均显著高于干扰组和对照组,实验组和干扰组BMP4蛋白在脂肪组织中的表达要显著低于对照组(均P<0.05)。结论:BMP4在肥胖小鼠体内心肌组织和动脉血管中明显高表达,是促进局部炎症反应发生的独立危险因子。Abstract: Objective:To investigate the bone morphogenetic protein 4(BMP4)expression in the myocardial tissue,aortas and adipose tissue of obese mice,and to analyze the correlation between BMP4 and inflammation.Method:The ob/ob mice were divided into experimental group and interference group,the C57BL/6mice were included in control group.There were 8mice in each group,fed with normal diet,and interference group received tail vein injection of shRNA BMP4 adenovirus,1times per week for 12 weeks.The samples of heart,abdominal aorta and abdominal adipose tissue took after 12 weeks,and the expression of BMP4 protein was detected by immunohistochemistry,the expressions of mRNA and protein of BMP4,IL-1βand IL-9were tested by quantitative PCR and Western blotting.Result:The mRNA and protein levels of BMP4,IL-1β,and IL-9were significantly higher in the myocardial tissue and aorta in experimental group than those in interference group and control group,while the levels of BMP4 protein in the adipose tissue in experimental group and interference group were significantly lower than those in control group.Conclusion:BMP4is significantly up-regulated in the myocardial tissue and aorta in obese mice,which is an independent risk factor involved in the local inflammatory response.
