摘要: 经典的基因表达理论认为,所有RNA分子都是线性结构,测序方法只能分离具有特征性分子尾巴的那些分子。环状RNA的末端连接在一起,缺乏这些尾巴,因此被普遍忽略掉了。所以环状RNA一直飞行于雷达之下,鲜被发觉。最新研究证明,环状RNA不仅存在,而且广泛存在于人体细胞内外,包括血液、唾液及心脏,提示环状RNA或可作为心血管疾病的新型治疗靶点和生物标记物。Abstract: Recently, circular RNAs (circRNAs) are identified as competing endogenous noncoding RNAs that sponge microRNAs or proteins by complementary base paring. The function of circRNAs is only now beginning to be understood. Unlike typical linear RNAs, the circRNAs lack the typical 5' cap and poly(A) tail. They are extraordinarily stable due to the lack of exposed terminal ends which are susceptible to degradation by exoribonucleases(RNases). They are present in whole blood, plasma and extracellular vesicles. The latest findings disclose that the circRNAs may constitute a novel reservoir of therapeutic targets and biomarkers of cardiovascular diseases.
