The expression of TRPA1in peripheral blood monocytes in patients with coronary heart disease and the relationship associated with the severity of coronary artery stenosis

  • 目的:通过检测瞬时受体电位离子通道A1(Transient receptor potential channel Al,TRPAl)蛋白及mRNA在冠心病患者外周血单核细胞中的表达,分析TRPA1 mRNA转录水平与Gensini积分的相关性。方法:根据冠状动脉(冠脉)造影结果将入院患者分为冠心病组(39例)和非冠心病组(36例),分离提取鉴定患者外周血单核细胞,通过Western blotting、RT-PCR分别检测两组患者单核细胞TRPA1的蛋白及mRNA表达水平。根据患者是否合并高血压、糖尿病,将冠心病患者分为高血压组与非高血压组、糖尿病组与非糖尿病组,分别比较两组间TRPA1 mRNA转录水平。分析冠心病患者单核细胞TRPA1 mRNA转录水平与冠脉病变Gensini积分的相关性。结果:①成功分离提取患者外周血单核细胞,经流式细胞术鉴定,单核细胞纯度在86.1%。②Western Blot显示,冠心病组患者单核细胞TRPA1蛋白表达水平高于非冠心病组(1.524±0.426∶0.590±0.275,P<0.01)。③RT-PCR显示,冠心病组TRPA1 mRNA转录水平高于非冠心病组(9.121±1.828∶3.923±2.535,P<0.01),高血压组TRPA1 mRNA转录水平高于非高血压组(9.812±1.180∶8.740±1.586,P<0.05),糖尿病组TRPA1 mRNA转录水平高于非糖尿病组(9.685±0.932∶8.982±0.756,P<0.05)。④冠心病患者TRPA1 mRNA转录水平与Gensini积分呈正相关(r=0.964,P<0.01)。结论:冠心病患者单核细胞TRPA1表达水平增高,TRPA1 mRNA转录水平可作为评价冠脉病变程度的新指标。
