The activation of T lymphocytes and expression of inflammatory indicators in young patients with essential hypertension

摘要: 目的:探讨年轻原发性高血压患者外周血T淋巴细胞活化状态及炎性因子表达水平。方法:收集36例年龄小于60岁的高血压患者及36例同年龄段的健康成年人的外周血,对所有研究对象均分离外周血单个核细胞,通过流式细胞术检测CD45RA+T淋巴细胞和CD45RO+T淋巴细胞的比例,ELISA法检测炎性因子白细胞介素(IL)-6、肿瘤坏死因子(TNF)-α的表达,分析CD45RA和CD45RO表达的相对关系,及炎性因子IL-6、TNF-α在年轻人群原发性高血压发病机制中的作用。结果:与健康对照组相比较,年轻原发性高血压患者外周血CD45RA+T淋巴细胞的比例显著降低[(42.85±10.62)%∶(33.72±12.40)%,P<0.01],而CD45RO+T淋巴细胞的比例显著升高[(57.16±10.62)%∶(66.28±12.40)%,P<0.01];炎性因子IL-6和TNF-α表达水平明显升高[(2.96±0.92) pg/mL∶(3.90±1.26) pg/mL,P<0.01;(2.25±0.39) pg/mL∶(2.56±0.49) pg/mL,P<0.01]。结论:外周血T淋巴细胞过度活化及炎性因子高水平表达,可能参与了年轻人群原发性高血压的发病。Abstract: Objective: To investigate the activation status of T lymphocytes and the expression of inflammatory indicators in peripheral blood of young patients with essential hypertension.Methods: Peripheral bloods of 36 hypertensive patients younger than 60 years old and 36 age-pared healthy adults were collected, and the mono-nuclear cells were separated from the peripheral of all subjects. The frequency of CD45 RA+T lymphocytes and CD45 RO+T lymphocytes were detected by flow cytometry. ELISA was used to detect the expression of inflammatory indicators IL-6 and TNF-α; and to analyze the relative relationship between CD45 RA and CD45 RO expression and the role of inflammatory indicators IL-6 and TNF-α in the pathogenesis of essential hypertension in young population.Results: Compared with healthy controls, the frequency of CD45 RA+T lymphocytes in peripheral blood of young patients with essential hypertension was significantly decreased[(42.85±10.62)% vs(33.72±12.40)%, P<0.01], while the frequency of CD45 RO+T lymphocytes was significantly increased[(57.16±10.62)% vs(66.28±12.40)%, P<0.01]. The expression levels of inflammatory indicators IL-6 and TNF-α were significantly increased[(2.96±0.92) pg/mL vs(3.90±1.26) pg/mL, P<0.01;(2.25±0.39) pg/mL vs(2.56±0.49) pg/mL, P<0.01].Conclusion: Excessive activation of T lymphocytes and high expression of inflammatory indicators in the peripheral blood may be involved in the pathogenesis of essential hypertension in young people.
